The following steps are usually followed when using a rotary evaporator (rotovap):
1. Prepare the sample: Load the sample into the round-bottom flask, not more than half the capacity of the flask.
2. Check the equipment: Make sure that all parts of the rotary evaporators are intact, including the flask, condenser and vacuum pump.
3. Install the flask: Install the round-bottom flask with the sample onto the rotary vacuum evaporator and fix it with a clamp.
4. Set the vacuum: Turn on the vacuum pump and adjust the vacuum degree by the control knob or switch on the vacuum rotary evaporator.
5. Heat the water bath: Add distilled water to the water bath and heat it to an appropriate temperature. Generally, the temperature of the water bath should be lower than the boiling point of the solvent.
6. Start rotating: Turn on the power of the rotary evaporator and start the motor to rotate the flask to increase the surface area of the sample and accelerate the evaporation process.
7. Evaporation process: The solvent evaporates under the conditions of rotation and heating, condenses after cooling through the condenser, and flows into the receiving bottle.
8. Monitor the process: During the evaporation process, the sample temperature and rotation speed need to be monitored to ensure that the sample does not overheat.
9. Complete evaporation: When the solvent has evaporated or the desired degree of evaporation has been reached, turn off the vacuum pump and stop heating and rotation.
10. Release the vacuum: Release the vacuum in the system by opening the valve between the vacuum pump and the rotary evaporator.
11. Remove the flask: Use the manual lift to remove the flask from the water bath and stop the rotation.
12. Clean the equipment: After the evaporation is completed, the condenser and receiving flask need to be cleaned and prepared for the next use.
